神経伝達物質であるドーパミンは、このたび50回目の誕生日を迎えました。1950年代後半にドーパミンが神経細胞として発見され、神経伝達の役割を果たすと考えられたことは、神経科学研究の分野では画期的な出来事でした。この画期的な発見は、脳を神経細胞の集合体としてとらえ、神経伝達物質の種類によって区別する時代の始まりであり、また、脳疾患の病巣を特定できるようになる重要な時代であった。中脳ドパミン系（mdDA）は、以前は中脳ドパミン系と呼ばれていたが、その発見以来、注目されている。ドーパミンが中枢神経系（CNS）の神経伝達物質であることが初めて明らかになり、精神・神経疾患との関連性が指摘されたことで、ドーパミンニューロンとその突起の機能に関する神経化学的、薬理学的、遺伝学的研究が数多く行われるようになった。この10年間、遺伝子発現や発生に関する研究により、この神経細胞集団に関する知識はさらに深まり、新たなレベルの複雑さが明らかにされた。mdDAシステムの分子的解明の始まりは、NurrlとPitx3のクローニングと特性評価であった。これらの転写因子は、mdDAの発生において重要な機能を持つことが示された。これらの初期研究に続いて、ドーパミン神経細胞許容領域の形成、前駆体の誘導、終末分化の誘導、そして最終的にはmdDA神経細胞プールの維持に重要な機能を持つ、他の多くのタンパク質が同定された。
